Here we report a case of testicular leiomyosarcoma occurring in a 32-weeks-old male senescence marker protein 30 (SMP30) knockout mouse. Grossly, a testicular mass completely replaced the right testis. The mass had a diameter of 1.6×0.9cm, and was well-encapsulated and firm. The cut surface showed heterogeneous white and yellow-brownish components with multifocal hemorrhages. Microscopically, the mass was composed of interlacing bundles of neoplastic cells characterized by a spindle-shaped appearance with elongated blunt-end nuclei and frequently observed mitotic figures. Immunohistochemically, the neoplastic cells showed strong positivity for vimentin and α-smooth muscle actin. Finally, present case was diagnosed as testicular leiomyosarcoma in an SMP30 knockout mouse. ©2017 PVJ. All rights reserved

Senescence marker protein 30 functions as gluconolactonase in L-ascorbic acid biosynthesis, and its knockout mice are prone to scurvy.
We originally identified senescence marker protein 30 (SMP30) as a distinctive protein whose expression decreases in an androgen-independent manner with aging. Here, we report its sequence homology found in two kinds of bacterial gluconolactonases (GNLs) by using the blast search. Then, through a biochemical study, we identify SMP30 as the lactone-hydrolyzing enzyme GNL of animal species. SMP30...

AIMS/INTRODUCTION Senescence marker protein-30 (SMP30) is abundantly expressed in renal proximal tubule cells, but its expression decreases with age. Previous studies have shown that reduced SMP30 expression could contribute to aging-associated deterioration of cellular function and tissue injury. In the present study, we investigated the effects of SMP30 deficiency on the pathogenesis of diabe...
متن کاملIncreased Susceptibility of Radiation-Induced Intestinal Apoptosis in SMP30 KO Mice
Recently, senescence marker protein-30 (SMP30) knockout (KO) mice have been reported to be susceptible to apoptosis, however, the role of SMP30 has not been characterized in the small intestine. The aim of the present study is to investigate the role of SMP30 in the process of spontaneous and γ-radiation-induced apoptosis in mouse small intestine. Eight-week-old male wild-type (WT) mice and SMP...
متن کاملStructural Basis of the γ-Lactone-Ring Formation in Ascorbic Acid Biosynthesis by the Senescence Marker Protein-30/Gluconolactonase
The senescence marker protein-30 (SMP30), which is also called regucalcin, exhibits gluconolactonase (GNL) activity. Biochemical and biological analyses revealed that SMP30/GNL catalyzes formation of the γ-lactone-ring of L-gulonate in the ascorbic acid biosynthesis pathway. The molecular basis of the γ-lactone formation, however, remains elusive due to the lack of structural information on SMP...
متن کاملPrimary testicular leiomyosarcoma.
Primary leiomyosarcoma of the testis is a rare condition with only 13 cases reported. We present a case of primary intra-testicular leiomyosarcoma in a 45 year old patient who presented with painless testicular enlargement. Ultrasound revealed a large heterogeneous right testicular solid mass with moderate hydrocele and huge retroperitoneal mass. Serum alpha-fetoprotein (AFP) and beta-human cho...
